CTSA Pharmaceutical Assets Portal

As a CTSA institution, the University of Michigan Medical School is able to participate in the CTSA Pharmaceutical Assets Portal – a project geared toward finding new uses for discontinued drugs.

The Portal project is jointly supported by the Pfizer Corporation and NCRR.  Portal members are given an unprecedented opportunity to request compounds from the Pfizer Indications Discovery Unit, a division of Pfizer vested with finding new uses for old drugs.  In addition to the compounds that you may be aware of from publicly-available sources, Pfizer is also encouraging inquiries about the existence/availability of little known Pfizer compounds that target specific mechanisms in which you may have an interest.  The ultimate goal is to leverage existing compounds to advance mechanistic understanding of human disease, resulting in novel treatments for patients.  Integration of academic investigators into collaborative repositioning efforts with Pfizer would substantially increase the knowledge base and the pool of methodologies available for proof of concept studies. These matches will undoubtedly result in an increased number of approved drugs for new indications and considerable public benefit.

Membership in the Portal is free for CTSA researchers. To learn more about the Portal project, visit the website. The website will also allow you the opportunity to complete a short survey describing your interests and to join the Portal.

If you have any questions, please contact Kate Marusina, Ph.D., M.B.A., Clinical and Translational Science Center, UC Davis School of Medicine, at 916-703-9177 or (kate.marusina@ucdmc.ucdavis.edu).

LSI's Innovation Partnership Accelerates Progress of Four Drug Discovery Projects in First Year

The Innovation Partnership at the Life Sciences Institute (LSI) marked its first year with one project spinning off into a new company focusing on obesity and diabetes treatment and the other three drug discovery projects making accelerated progress towards treatments in the areas of neurodegenerative disease, bacterial infection, and cancer. The first-year results are detailed in a report just issued by the LSI.

The spinoff, Vega Therapeutics, has licensed technology from the lab of LSI director Alan Saltiel. Saltiel’s project was based upon his discovery of the role of the IKKE gene in obesity and diabetes. The Innovation Partnership supported the development of that discovery to the point where investors were interested in starting a company around it.

The Innovation Partnership is a scientific discovery enterprise that offers LSI scientists targeted funds and mentorship to focus on projects that might normally fall by the wayside into the gap in the drug discovery pipeline traditionally known as the “valley of death.”

As a scientist who received funding in the Innovation Partnership’s first year, Jason Gestwicki, Ph.D., describes its impact: “Getting an experienced mentor group in place who can see the entire trajectory of your project and know what people and resources are needed to get your discovery from the bench to the bedside is so valuable. They can put the pieces in place to get the process moving in the right direction.”

Since its inception in 2009, the Partnership has awarded $635,000 in gift funds from individuals and businesses to support the projects.

U-M Receives NIH, FDA Grant to Study Adaptive Clinical Trial Designs

Researchers hope innovative design will accelerate drug, device evaluation, improve safety for patients, and improve efficiency, costs of clinical research.

Clinical trials in which participants are randomly assigned to different treatments by design are the cornerstone of clinical research. When conducted correctly, it’s widely accepted by the research community that the results are valid and can be trusted.

But researchers at the University of Michigan hope to improve the process for bringing therapies and medical devices to patients by investigating the impact of a class of innovative trial designs known as adaptive clinical trials – which make adjustments to the trial using information accumulated as patients are enrolled – in hopes of improving the efficiency of clinical trials as a whole.

“One of the hard and firm rules of randomized clinical trials, and for good reason, is you can’t just go changing your trial to get the results you want,” says William Barsan, M.D., Chair and Director of the U-M Department of Emergency Medicine.  “But with adaptive clinical trials, you spend a lot of time up-front asking yourself, ‘What if?’ so that any changes made throughout the course of the study are all agreed upon at the beginning, written into the study design and only changed throughout the course of the study based on pre-specified rules.”

Researchers cannot always predict at what point a specific drug or treatment will show therapeutic results during the course of the study, if at all. Once a standard trial is underway, trial characteristics such as inclusion criteria cannot be modified or changed even if a sufficient amount of data collected before the trial’s end-date demonstrates that such changes may be necessary.

For example, remaining patients who are randomized to the placebo group cannot benefit from the therapeutic effect of the treatment, and money will continue to be spent until the trial ends. Current trials often include periodic review of data to search for early benefit or early harm but changes are usually only triggered when an overwhelming effect is seen.

Adaptive clinical trials can more seamlessly make use of accumulating information using Bayesian statistics – which allow data analysts to predict trial success based on early patient responses and other accumulating information.

“Adaptive design essentially allows the statisticians to continuously reanalyze the data over the entire study, and we can end up changing how patients are assigned within the study if it really looks like it’s working,” says Barsan, who is also principal investigator of the clinical trials coordinating center of the Neurological Emergencies Treatment Trials Network. “We believe this is one way to avoid getting false negatives and it‘s better protection for patients which is really important.”

The three-year project, “Accelerating Drug and Device Evaluation through Innovative Clinical Trial Design,” is one of four recipients of a $9.4 million award by the National Institutes of Health-Food and Drug Administration Joint Leadership Council to spearhead collaborative activities to stimulate a new research in regulatory science.

The project is led by Barsan, Donald A. Berry, Ph.D., Senior Statistical Scientist and founder of Berry Consultants, and Roger J. Lewis, M.D., Ph.D., Co-chair of the Regulatory and Ethics Knowledge and Research Program and Professor of Emergency Medicine at the University of California –Los Angeles.

The objective of the grant is to “optimize the design of four large, neurological emergency trials at various stages of development,” Barsan says. “We’ll use modeling and mathematical simulation to really kick the tires on a number of potential adaptive designs with the hope of gaining efficiency in the ultimate trial design. Hopefully it will lead to considerable savings in the research process and allow us to more accurately and rapidly identify treatments which improve patient outcomes.”

Animal Research

UCUCA USDA Annual Report

September 30, 2010 was the end of the USDA's reporting year.  The Annual Report of Research Facilities for the period October 1, 2009 thru September 30, 2010 is due to the USDA on December 1, 2010.

Contained in the USDA Report will be the number of animals per species born, acquired, or used by UM during the reporting year and detailed explanations of animal use involving pain, discomfort, or distress for which pain-relieving drugs could not be used because they would interfere with the experimental result.

Please help us compile a complete and accurate USDA report!  Here's how:

They should go back as far as October 1, 2009, if necessary.  This applies primarily to researchers breeding their own animals or capturing animals, but could also apply to animals acquired from other institutions, offspring of animals acquired pregnant, and animals purchased by the litter rather than individuals.  If researchers purchase their animals through the ULAM Business office, or use ULAM barcoding, the numbers of animals purchased/born are gathered automatically.

  • Report Carryover Animals

If there were any animals in your area/laboratory during a significant portion of this reporting year (October 2009 – September 2010) that were acquired MORE than a year earlier (i.e., animals which remain at the University for more than one year), please send these numbers to us via email, or call Monika at 734-615-7032.  We’ll need to know the principal investigator, species, protocol number, and number of animals per Use Category.

If you ever have any questions regarding the UCUCA or anything else related to your animal work or compliance, please do not hesitate to contact the UCUCA Office (ucuca.office@umich.edu or 734-763-8028).
