Research Description:

1) A Role of Stem Cell Factor (SCF) in Chronic Asthma, funded by the NIH, PI Nicholas W. Lukacs: We hypothesize that SCF production during allergic airway responses augments the progression of disease via eosinophil degranulation and production of profibrotic mediators leading to detrimental pathophysiology and airway remodeling. Blocking SCF-mediated activation within the airway using a monoclonal antibody indicates that this cytokine may represent a viable target for therapeutic intervention that could affect multiple aspects of allergen-induced immunopathology. The goal of project is to determine SCF signaling leading to eosinophil activation and specific profibrotic cytokine production. In addition, changes in airway physiology in normal and SCF deficient allergic mice would be assessed to elucidate the mechanism of SCF-induced eosinophil activation during disease development.

2) Bone marrow progenitor cells in lung fibrosis, funded by the Sandler Program for Asthma Research, PI Sem H. Phan, Co-PI Nicholas W. Lukacs. The goals of the project is to investigate a) whether fibroblast-like collagen producing cells involved in airway remodeling during chronic asthma may originate from bone marrow; b) whether previously observed beneficial effects of anti-SCF treatment on chronic asthma is due to the suppressed trafficking of bone marrow derived cells into the sites of airway remodeling.
